If she invited you to church and makes a clear point to sit next to you then she must like you and enjoy being around you. especially if she put the time and thought into getting you a card and gift and then put even more effort in writing a birthday wish on it i would say she is definitely interested. A girl won't go through all that trouble for just anyone.
